How Dog DNA Tests Work: Swab, Lab, and Report Explained

How a dog DNA test goes from cheek swab to breed-and-health report. SNP genotyping, breed reference panels, and what 2 to 4 weeks of lab work actually does.

A dog DNA test looks simple from the outside: swab the inside of your dog’s cheek, mail it back, get a report. Under the hood, three things happen between the swab and the report, and understanding them helps you read the results properly.

Step 1: sample collection

The kit ships a sterile cheek swab. You rub it against the inside of your dog’s cheek for around 30 to 60 seconds to collect cells. No blood, no veterinary visit. The cells include enough DNA to run the lab work, assuming your dog has not just eaten or drunk water in the previous half hour.

You seal the swab, drop it in the prepaid mailer, and the kit travels to the company’s lab. Both Embark and Wisdom Panel use this same basic collection method.

Step 2: SNP genotyping at the lab

Both major dog DNA tests use SNP genotyping rather than whole-genome sequencing. The lab extracts DNA from the swab cells and reads it across a custom array of hundreds of thousands of single-nucleotide polymorphism sites. These are specific positions in the genome where individuals commonly differ.

Embark reads across 200,000+ markers. Wisdom Panel reads across a comparable array. Both arrays are chosen specifically to capture variation that is informative for canine breed differentiation and for known disease-associated variants.

The result is a long file of genotype calls, one per marker: which version of the variant your dog carries at that position.

Step 3: breed reference panel comparison

This is the part the report calls “breed identification.” The company has already built a reference panel by genotyping dozens to hundreds of confirmed purebred dogs of every recognized breed. Your dog’s genotype file is then compared against these reference profiles.

The algorithm scans segments of your dog’s genome and asks, for each segment, which breed reference profile it most closely matches. The percentages on your report are the sum of these segment assignments, weighted by confidence. A pure-breed dog will match overwhelmingly to one reference profile. A mixed-breed dog’s genome gets carved up across multiple references.

Step 4: health marker panel

A separate analysis checks specific positions in your dog’s genome against a panel of variants known to be associated with inherited diseases. Embark screens 250+ conditions; Wisdom Panel Premium screens 210+. Each condition has one or more specific marker positions; the test reports whether your dog carries 0, 1, or 2 copies of the relevant variant.

The output is per-condition: clear at one of the variants, carrier (one copy, no expected symptoms but can pass to offspring), or at-risk (two copies of a recessive variant, or one of a dominant variant). For the deeper explainer, see dog DNA health screening explained.

Critical: a flagged result is screening context, not a veterinary diagnosis. A “carrier” or “at-risk” result is a reason to talk to your vet about the specific condition, not a confirmation of disease.

Step 5: report generation and timeline

The company assembles all of this into a readable report: breed pie chart, trait predictions, health condition list, and any extras like Embark’s Relative Finder (which compares your dog’s genotype against every other Embark-tested dog to find shared segments).

Total turnaround is 2 to 4 weeks at Embark, 2 to 3 weeks at Wisdom Panel, measured from sample receipt at the lab. Shipping the kit there adds 2 to 5 business days on the front end.

What the test does not do

It does not sequence your dog’s whole genome; both major tests read a targeted array of markers. It does not predict diseases that are not on the panel, and it does not catch breed-specific risks that lack a clean single-marker test. It does not provide a clinical diagnosis. And it does not detect mosaic conditions, structural variants, or epigenetic factors.

For the purpose it is designed for, identifying breed ancestry and screening for known inherited markers, the test does a credible job. For anything beyond that, you need a veterinarian.
