mtDNA Testing and Maternal Lines: What It Reveals
How mitochondrial DNA testing traces the strictly maternal line, what the mtFull Sequence at FamilyTreeDNA delivers, and when this test is worth ordering.
Mitochondrial DNA, or mtDNA, is the small loop of genetic material inside mitochondria, the energy organelles inside almost every cell. Every person inherits mtDNA exclusively from their mother. Fathers do not pass theirs on. That single inheritance pattern is what makes mtDNA testing a tool for tracing the strictly maternal line: your mother, her mother, her mother, and so on back through deep time.
Both men and women inherit mtDNA, so this test is available to anyone. What it cannot do is tell you about any line other than the direct maternal one. The maternal grandmother on your father’s side, for example, contributed no mtDNA to you.
What you get from a consumer mtDNA test
The standard consumer option is the mtFull Sequence test from FamilyTreeDNA, priced around the higher end of their kit range as of early 2026. It reads all 16,569 base pairs of the mitochondrial genome and returns:
- Your mtDNA haplogroup at full resolution
- A list of mtDNA matches in the FamilyTreeDNA database
- A list of every variant you carry compared to the reference sequence
The full sequence is what most genealogists buy now; older partial tests (HVR1 and HVR2 only) are still around but produce far less actionable match data.
Why mtDNA is slower and deeper than autosomal
Mitochondrial DNA mutates much more slowly than the nuclear DNA tested by autosomal kits. Two people with the same full mtDNA sequence might share a common maternal-line ancestor a few hundred years ago, or several thousand years ago. mtDNA is poor at identifying close, named relatives, and excellent at placing your maternal line into a deep historical and geographic context.
That trade-off is the entire reason to order it. If you want to know who your third cousins are, AncestryDNA and the other autosomal services are the right tool. If you want to know which deep maternal lineage your maternal line descends from, mtDNA is the only consumer test that answers the question.
Haplogroups, briefly
An mtDNA haplogroup is a label for a branch of the global maternal tree. Haplogroup H is the most common in Western Europe. Haplogroup U includes some of the oldest lineages in Europe. Haplogroup L groups originated in Africa and remain most diverse there. M and N cover much of Asia. Our haplogroup explainer walks through the classification system.
Knowing your haplogroup does not tell you a nationality or an ethnicity. It tells you which deep branch of human migration history your direct maternal line sits on. National Geographic’s Genographic Project popularised the framing, and MITOMAP and PhyloTree are the academic references most testers consult.
Where the matches help
Two testers with identical or near-identical full mtDNA sequences share a maternal-line ancestor more recently than two random testers, but recently in mtDNA terms can still mean centuries. Where matches do pay off is when combined with a documentary tree. If you and a match both descend from the same village or surname cluster on the maternal line, an identical mtFull match supports that connection in a way autosomal data sometimes cannot at that distance.
Living DNA also reports an mtDNA haplogroup as part of its kit, but without the full-sequence matching database that FamilyTreeDNA offers.
Limits to keep in mind
The mtDNA reference panel does not assign a country. Names like “the Seven Daughters of Eve” are popular-science framing rather than science itself. Haplogroup labels evolve as more sequences are added to the reference tree; expect your label to gain detail (extra letters or numbers) over time.
For most people building a family tree, autosomal testing is the better first purchase. mtDNA testing earns its price when you have a specific maternal-line question that the other tests cannot answer.
