DNA Test Sample Collection Guide: How to Avoid a Failed Kit

A practical step-by-step guide to collecting saliva or cheek-swab DNA samples correctly, the most common errors that fail a kit, and what to do if yours does.

A failed DNA sample is the single most preventable delay in the whole process. The lab work itself runs on a predictable schedule. The part most buyers stumble on is the ten minutes at the kitchen sink, where small mistakes can mean your kit gets returned as “insufficient DNA” and the four-to-six-week clock starts over.

Register your kit before you spit

Every major brand ties your sample to a unique barcode printed on the collection tube and registration card. If you mail a tube without registering the barcode to your account, the lab cannot match your sample to a person and the kit is effectively useless. Register at the URL printed on the box (typically the merchant’s activation page) before you open the tube.

This step takes two minutes and is where roughly half of all “lost kit” complaints actually originate.

Saliva tubes: the 30-minute rule

AncestryDNA, 23andMe, and MyHeritage all use saliva collection tubes. You spit into the tube up to a fill line, screw on a cap that releases a stabilizing fluid, and shake to mix.

The single most important rule is the 30-minute exclusion window before collection:

  • No food
  • No drink (including water in some protocols; check your kit insert)
  • No gum, mints, lozenges, or tobacco
  • No teeth brushing or mouthwash

Residual food, drink, or cosmetic mouth products dilute and contaminate the sample, which is the most common reason a lab requests a re-collection. Set a timer, then collect.

A few practical tips that help:

  • Rub your cheeks gently from the outside before you start. This boosts saliva flow without contaminating the sample.
  • Spit, do not blow bubbles. The lab needs cell-rich saliva, not foam. Most kits ask for 2 to 3 ml of actual liquid below the bubble line, which is more than first-timers expect.
  • Take your time. Five to ten minutes of patient collection is normal.

Cheek swabs: rub firmly for the full time

FamilyTreeDNA and many pet DNA tests use cheek-swab collection rather than saliva tubes. A swab is a small plastic-handled brush you scrape against the inside of the cheek for the full time the instructions specify, usually 30 to 60 seconds per swab.

Two things make swab collections fail:

  • Not rubbing firmly enough. The goal is to scrape loose cells from the cheek lining, not to mop saliva. Press the brush against the inside of the cheek and rotate.
  • Not waiting the full time. A ten-second swab will not give the lab enough material.

Let the swabs air-dry as the instructions direct before sealing them in the return envelope. Sealing damp swabs encourages bacterial growth that can degrade the DNA.

Special situations

If you have dry mouth, are dehydrated, or take medication that reduces saliva, give yourself extra time and consider hydrating well an hour before (then stopping water in the 30-minute window). For young children and many older adults, a cheek-swab kit is usually easier to complete than a saliva tube.

The mouth’s bacterial population does not invalidate a sample on its own. Modern labs separate human DNA from bacterial DNA during processing.

What if your sample fails

A “no result” or “insufficient DNA” notice is common and is not a reflection on your genome. Every major brand will send a replacement kit at no extra cost on the first failure. Contact the merchant’s support line, reference your kit ID, and request a replacement. AncestryDNA, MyHeritage, and FamilyTreeDNA all publish this policy openly.

Repeat the collection following the rules above. The replacement almost always processes successfully.
